There's not much slack in the line so you will want it unhooked before the trans gets separated from the motor. Otherwise you'll end up ripping it out and having to replace it.
Like I said, find the clip at the line coupler and pry it up about 1/2" to the 'release' position with a small screwdriver and then the line will pull out of the fitting. You will still have some resistance from the rubber O ring but it will pop out. If you completely remove the metal clip then don't lose it, and put it back on before reinstalling the transmission.
